Determination of proteins on surfaces
This datasheet describes the determination of Arginine,
Ovalbumin and Human IgG on surfaces by a swabbing and fluorescence
method, using a portable spectrofluorimeter.
Introduction
The SMF 2 instrument is designed to measure trace levels of fluorescent
chemicals directly on surfaces for industrial hygiene, cleaning
efficiency measurement and quality control. It is fully portable,
produces a spectral output so that quantification is not solely
dependant on single wavelength readings and has data logging capabilities.
It is anticipated that these methods will be of interest to companies
operating biomanufacturing fermenters, those who process proteins
in pharmaceutical plant and any organization which needs to measure
and control these compounds on surfaces.
Analytical methods
Water solutions of proteins and amino acids were pipetted directly
onto filter paper for method development, or onto glass slides
for simulation of a surface monitoring activity. Samples of 1-15
microgram of the protein were reacted with 15 microlitres of 1%
triethanolamine and 15 microlitres of 0.2g/L Fluoram solution,
either on the filter paper for method development or onto the paper
wipe. Each yielded a fluorophor which was stable for several hours
thus allowing efficient preparation of multiple samples and their
measurement.
Readings were made after a drying and colour development period
of 20 minutes had elapsed, with the sample held in the dark. |
